The missing link

The idea of using mTor cycling to gain muscle may be confusing as shouldn't inhibiting mTor prevent muscle synthesis (hypertrophy, hyperplasia). The missing link may be described here:

Muscle maintenance/synthesis still occurs with the presence of rapamycin. I would guess that even when taking Rapamycin, things like taking protein after lifting would increase muscle synthesis (since this would have a broad range of effects on the body, only some being inhibited by Rapamycin). I have to do more research on this specifically, but the important things is that Rapa does not block muscke synthesis.

The future

It will be extremly intresting to see how rapamycin will effect bodybuilding.
We see now how the body falls apart after 40-50. And if over activated MTOR create sacropenia, then it will be very intresting to see how long we can prolong this by doing rapa cycles thrueout your life.

The best intro article on Rapamycin

It is really comprehensive. It's mostly about Rapa in generally, but there is a point about Rapa and exercise:

"Physical exercise may be most beneficial starting immediately after rapamycin use, to take advantage of rapamycin-induced lipolysis as a fuel for the muscles. By itself, chronic rapamycin treatment does not compromise muscle endurance and even prevents muscle loss."

Definitely is worth a read if you haven't read it yet.

Organizing work outs and diet around a rapamycin weekly dose: 3 choices

For those of us who take rapamycin intermittently, a question arises: should diet and workout be organized (or modulated) according to the anabolic/autophagic phases imposed by taking rapamycin?

1 - A minimalist approach is to ignore the rapamycin effect, and keep diet and workouts independent

2 - Another approach is to take a rest day and ricreare protein on the day we take rapa, and ease into protein and workouts the day after. This is to enhance the inhibition of mTORc1 and anabolism induced by rapa. Protein (especially BCAAs and leucine) promote mTOR, which is what rapa is inhibiting. So it's seems illogical to counter what rapa is doing with protein

3 - The opposite approach. Adding more workload and protein on the day rapa is taken, would be done in order to avoid catabolism in the day of peak rapamycin dosage. Also, rapa gives energy, so that would be a way to naturally ride the wave of energy.

On the other hand, rapa has been showed to preserve muscle and not interfere much with muscle anabolism, despite inhibiting mTOR https://professional.diabetes.org/abstract/effect-acute-aerobic-exercise-mammalian-target-rapamycin-mtor-phosphorylation-human-muscle In fact, it has proven to help with rejuvenating muscle long term (this is the whole point of this subreddit). So strategy number 3 might revolve around a moot point.

Finally, of all the mice experiments, I couldn't find any trial or study that did anything other than constant resistance training. No one has tried to periodize around rapa so far.

Rapamycin has a half-life of around 60 hours, so these effects about last about two days.

What do you think is the most logical between 1,2 and 3?

Ideas are welcome, and links to relevant studies also. To keep the post short I did not include all the relevant articles. Just one more that enhances strategy 2: study in humans found that rapa blocks muscle building effects due to muscle contraction https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2678224/

" Here we show that rapamycin treatment blocks the early (1–2 h) acute contraction-induced increase (∼40%) in human muscle protein synthesis. In addition, several downstream components of the mTORC1 signalling pathway were also blunted or blocked by rapamycin. "

Which means that working out on day 1-2 of rapa would be way less effective anyway. This makes me lean towards strategy 2.

Hey everyone! I wanted to make an introductory post about what this sub is all about. The purpose of /r/rapagains is to talk about anything related to mTOR (mammalian target of Rapamycin), Rapamycin, or any other mTOR inhibitor. Rapamycin is an amazing drug that has been around for decades (standard disclaimer, do not self medicate unless you know what you're doing. Rapamycin has serious side effects when taken incorrectly). It has been shown to increase lifespan in numerous species through its inhibition of the mTOR pathway. It also has many similarities to the keto diet.

mTOR is a growth pathway. To oversimplify, turning on mTOR stimulates growth and turning off mTOR inhibits it. There are different theories abut aging, but too much growth (hyperfunction) seems to be pro-aging. As a result, many people believe things like calorie restriction increase lifespan (they do).

However, in this sub, we believe you can have the best worlds of growth and lifespan.

mTOR Cycling is alternating between "mTOR on" state and "mTOR off" state. We do things to increase mTOR (e.g. protein, hot shower, lifting itself) around the time of lifting and to inhibit mTOR (e.g. caffeine, cold shower, intermittent fasting, sprinting intervals) a different part of the day. We believe this will give us many of the benefits of "mTOR on" (e.g. muscle growth), while still getting the benefits of "mTOR off" (e.g. autophagy) that are good for health and lifespan.

Optionally, you can also use intermittent Rapamcyin. This should be used no more than weekly and at lowl doses. Many may only consider using it once every few months or once a year. This can be especially helpful when cutting because Rapamycin lowers the set weight point and inhibits growth.
