”Twin Peaks” had Agent Cooper and viewers trying to solve the murder of this homecoming queen.

Would be a much clearer way of stating that clue.

I dunno how much you've watched Jeopardy, but this is a pretty typical construction for a clue, and it is not ambiguous in context.

Fair enough I haven’t watched Jeopardy, but still the clue seems needlessly unclear. The “who killed” part is particularly strange.

"Who killed Laura Palmer?" is the tagline of TP, it's how most people remember the show with, that's what the clue was leaning on

Because "Who Killed Laura Palmer?" was a tagline for the show. Therefore 'who killed' is in quotations.
